Unlike EBV immortalized B cells, SV40 large T antigen immortalization can be applied to a wide range of cell types, but has lower specificity for B cells. On the contrary, EBV immortalization is specifically targeted at B cells, utilizing natural virus host interactions to achieve immortality.

 

EBV Immortalized B cells:

 

Immortalized B cells by EBV is an effective procedure for inducing the long-term growth of certain B cells. The process can be divided into three stages: virus preparation, preparation of the target cells to be immortalized, and EBV infection and growth of infected cells. Infection of B cells with EBV gives rise to growth-transformed and immortalized lymphoblastoid cell lines (LCL) in vitro. Among their many applications is the use of LCL to present antigens in a variety of immunologic assays and to generate human monoclonal antibodies.

The methods for producing monoclonal antibodies in immortalized B cell line include hybridoma technology, phage display, and B cell cloning: directly cloning antigen-specific B cells from immunized animals or humans, and immortalizing them to express specific antibodies using techniques such as EBV transformation or CRISPR mediated gene editing.

EBV's hosts are highly targeted, which tend to infect only B cells in humans and certain primates, limiting its use in other species. KMD Bioscience has successfully immortalized B cells derived from humans, rats, mice and pigs by using this technology.

Immortalized B Cells From Different Sources :

 

1. Human B Cell Immortalization

The most common method for human b cell immortalization involves the use of EBV, which naturally infects B cells and transforms them into continuously proliferating lymphoblastoid cell lines (LCLs).

--Process: Peripheral blood mononuclear cells (PBMCs) are isolated from blood samples, and the B cells among them are exposed to EBV, typically from the B95-8 cell line or similar. Following infection, cells are cultured in conditions that promote their transformation and proliferation.

2. Rabbit B Cell Immortalization

Rabbit B cells can also be immortalized, but the process is less straightforward due to the species-specific nature of EBV. Researchers have experimented with EBV-like viruses from rabbits or using similar techniques involving transfection with genes that promote cell growth and inhibit apoptosis.

- Process: Techniques may include the use of myc genes or other oncogenes to transform rabbit B cells. Alternatively, rabbit-specific viruses that function similarly to EBV in humans have been explored for immortalizing rabbit B cells.

- Applications: Rabbit B Cell Immortalization is valuable for producing polyclonal and monoclonal antibodies, especially for antigens where the rabbit immune system offers advantages over murine systems.

3. Immortalization of Primary B Cells

- Process: Primary B cells, isolated from blood or tissue samples, are exposed to EBV in vitro. The virus induces the cells to enter a state of continuous growth, resulting in immortalized cell lines. Careful culture conditions, including the use of feeder cells or cytokines (like IL-4), can enhance the efficiency of immortalization and the growth of the resultant cell lines.
