CD276/B7-H3 Target

CD276 (Gene ID:80381), also known as B7-H3, plays a crucial role in the immune system and has been extensively investigated for its involvement in cancer. It encodes a transmembrane protein of type I and is a member of the B7 family of immune checkpoint molecules. This protein has two immunoglobulin structural domains in mice, but due to exon duplications, it has four immunoglobulin structural domains in humans.

 

The primary function of CD276 is to participate in the regulation of T cell-mediated immune responses. It exerts a protective effect on tumor cells by inhibiting natural killer cell-mediated cytolysis and serves as a marker for the detection of neuroblastoma cells. CD276 is also involved in acute and chronic transplant rejection, the regulation of mucosal surface lymphocyte activity, and the creation of an immune milieu for the placenta and the fetus throughout gestation. Both isoforms of CD276 are redundant in their ability to regulate CD4 T cell responses. Specifically, isoform 2 enhances the induction of cytotoxic T cells and selectively stimulates interferon-γ production in the presence of T cell receptor signaling.

 

CD276/B7-H3 is commonly expressed in both normal tissues and solid tumors, but its protein is preferentially expressed in tumor tissues. The 3'UTR of its transcript contains the target site of miR29 microRNA, indicating that its expression is regulated by miR29. CD276 has been found to be aberrantly expressed in various types of cancer, suggesting its involvement in tumorigenesis. It acts as an immune checkpoint molecule in the epithelial-mesenchymal transition (EMT) pathway and plays a crucial role in cell proliferation, invasion, and migration, thereby positioning it as a potential therapeutic target for malignant tumors.
