TNFRSF17/BCMA Target

TNFRSF17 (tumor necrosis factor receptor superfamily member 17) (Gene ID: 608), also known as BCMA (B-cell maturation antigen), is a gene encoding a cell surface receptor expressed primarily on B cells and plasma cells. It plays a pivotal role in the regulation of B cell maturation, survival and antibody production. TNFRSF17 was initially identified as a receptor selectively expressed on mature B cells and plasma cells. Its role in B cell biology and antibody production has garnered significant research interest, particularly in the context of immune response and multiple myeloma. The functions of TNFRSF17 are as follows.

 

1. B-cell maturation: TNFRSF17 is essential for the maturation of B-cell in the bone marrow and secondary lymphoid organs, facilitating their differentiation into plasma cells responsible for antibody production.

 

2. Survival Factor: TNFRSF17 signaling confers survival signals to plasma cells, thereby extending their lifespan and ensuring continued antibody production. This is essential for the maintenance of immune response and humoral immunity.

 

3. Antibody production: The binding of TNFRSF17 to its ligands B-cell activating factor (BAFF) and proliferation-inducing ligand (APRIL) is required for optimal antibody production by plasma cells. Dysregulation of this signaling pathway leads to immunodeficiency.

 

4. Multiple myeloma: TNFRSF17 has been implicated in the pathogenesis of multiple myeloma, a malignancy of plasma cells. In multiple myeloma, malignant plasma cells often aberrant expression TNFRSF17, leading to uncontrolled cell proliferation and antibody production.

 

5. Therapeutic targets: Due to its involvement in the pathogenesis of multiple myeloma and autoimmune diseases, TNFRSF17 has been extensively investigated as a promising therapeutic target. Monoclonal antibodies targeting TNFRSF17, such as belantamab mafodotin, have been developed for the treatment of multiple myeloma.
