Tie-2, also known as TEK (tyrosine kinase with immunoglobulin-like and epidermal growth factor homology structural domain 2) (Gene ID:7010), encodes a receptor protein expressed predominantly on endothelial cells lining blood vessels. Tie-2 plays a key role in angiogenesis, the process of forming new blood vessels, and vascular homeostasis. It is considered to be a receptor tyrosine kinase involved in vascular development. Belonging to the Tie receptor family, which includes Tie-1 and Tie-2, extensive research has been conducted on Tie-2 due to its significance in a variety of physiological and pathological processes. The primary functions of Tie-2 are as follows.
1. Angiogenesis: Tie-2 is a key regulator of angiogenesis. It is activated by its ligand, angiopoietin-1 (Ang-1), which promotes vascular stabilization and maturation. 2. Vascular integrity: The activation of Tie-2 contributes to the maintenance of vascular integrity by strengthening endothelial cell-cell junctions and reducing vascular leakage. This function is essential to prevent excessive bleeding and edema.
3. Inflammatory response: Tie-2 signaling regulates the immune response and inflammation. Activation of Tie-2 reduces the expression of endothelial cell adhesion molecules and inhibits leukocyte adhesion, which contributes to the resolution of inflammation.
4. Tumor angiogenesis: The dysregulation of Tie-2 signaling, which promotes tumor angiogenesis, growth and metastasis, has been observed in various cancers. Targeting Tie-2 signaling is considered a potential anti-cancer therapy.
