ILT3, also known as LILRB4 (leukocyte immunoglobulin-like receptor B4) (Gene ID: 11006), encodes a cell surface receptor belonging to the immunoglobulin-like transcript (ILT) family. It is predominantly expressed on immune cells and plays a role in immune regulation and tolerance. ILT3 is a member of the ILT family, which consists of receptors that have immune regulatory receptors with immunomodulatory functions. These receptors are expressed on immune cells and interact with antigen-presenting cells and tissue-associated molecules. As an inhibitory receptor, ILT3 is expressed on dendritic cells (DCs), monocytes, and other immune cells, and interacts with ligands such as major histocompatibility complex (MHC) class i molecules and nonclassical MHC molecules (HLA-G). This interaction transmits inhibitory signals that regulate the immune response. The expression of ILT3 on dendritic cells (DCs) regulates antigen-presenting cells and T-cell activation. ILT3 can inhibit dendritic cell maturation by reacting with its ligands on dendritic cells, leading to reduced T-cell activation, which may contribute to immune tolerance and prevent over-immune responses to self-antigens. ILT3 is involved in immune tolerance mechanisms, and its interaction with HLA-G is associated with immune tolerance and may play a role in protecting fetal tissues from maternal immune responses during pregnancy. In the context of cancer, certain tumor cells can upregulate HLA-G and engage ILT3 on immune cells to suppress anti-tumor immune responses. This interaction contributes to immune evasion of tumors.
