INDO, also known as IDO1 (indoleamine 2,3-dioxygenase 1) (Gene ID:3620), is a gene encoding an enzyme called indoleamine 2,3-dioxygenase 1. This enzyme plays a crucial role in the regulation of the immune system and has been implicated in various physiological and pathological processes. Indoleamine 2,3-dioxygenase 1 is involved in tryptophan metabolism, an essential amino acid, and is widely expressed in various tissues, such as the placenta, immune cells, and some tumors. The significance of indoleamine 2,3-dioxygenase 1 lies in its involvement with immune tolerance and its potential as a target for cancer immunotherapy. It acts as a key regulator by catalyzing the breakdown of tryptophan into kynurenine and other metabolites within the tryptophan degradation pathway. Reduction of tryptophan and accumulation of kynurenine inhibits T cell activation and proliferation and contributes to immune tolerance. IDO1 is usually upregulated in the tumor microenvironment. Tumor cells and antigen-presenting cells can express IDO1, leading to immune evasion by inhibiting cytotoxic T cell activity while promoting the development of regulatory T cells (Treg). This promotes tumor growth and metastasis. IDO1 can be induced during infection as part of the host defense mechanism. It can help limit the growth of certain pathogens by limiting the availability of tryptophan. Dysregulation of IDO1 has been associated with autoimmune diseases. In some cases, increased IDO1 activity may result in immunosuppression leading to autoimmune diseases development.
